The Potential Use of Elastic Tissue Autofluorescence in Formalin-fixed Paraffin-embedded Skin Biopsies

Autofluorescence (AF) or naïve-florescence is the natural emission of light by biomolecules. During florescence microscope examination, we realized that elastic tissue brighter more autoflourescent than collagen and other biomolecules/cells in skin. Consequently, decided to review tissue-related pathologies under a report possible benefits this technique from selected cases paraffin-block archive, using protease digestion immunofluorescence method. Selected clinic-pathologically confirmed 3 elastofibroma dorsi, pseudoxanthoma elasticum, anetoderma, arteriovenous malformations, temporal arteritis, scar highly solar-damaged samples skin 2014-2019 were retrieved. Under fluorescent microscope, coarse, thick globularly-fragmented fibers shortened, irregular convoluted internal membranes arteries their integrity was visualized. None anetoderma had any signal representing tissue. It shown can be observed easily fluorescence case FFPE tissues. The resulting autofluorescence useful recognizing pathologies, it may used as an ancillary alternative method routine histochemical techniques.
